There are 2 possible ways:
1) use the native TPT ui, see https://powdertoy.co.uk/Wiki/W/Lua_API:Interface.html
2) register a mouseclick function, and whenever the mouse pointer is within your rectangle, just return false, which will tell TPT to ignore the click
